'Soft-particle' analysis of the electrophoretic mobility of a fibrillated and non-fibrillated oral streptococcal strain: Streptococcus salivarius.

چکیده
The electrophoretic mobility of microbial cell surfaces can be analysed in terms of a so-called soft layer model, in which the electrophoretic mobility is described as originating from the potentials over the surface charge layer and the membrane fixed charges. Often, the polyelectrolyte layer deforms under the influence of ionic strength variations. In the soft layer analysis of electrophoretic mobilities this is expressed in the softness 1/lambda. Here, we determined the softness of two oral streptococcal strains, S. salivarius HB and HBC12 from particulate microelectrophoresis in KCl solutions of varying ionic strength. Electron microscopy of negatively-stained organisms and X-ray photoelectron spectroscopy showed that strain HB had several classes of proteinaceous fibrils with lengths up to 178 nm on its outermost surface, while variant HBC12 had a bald, peptidoglycan-rich outer surface. The fibrillated strain HB appeared as relatively soft (1/lambda equals 1.4 nm) from analysis of its electrophoretic mobility, while the bald variant HBC12 was hard (1/lambda equals 0.7 nm) due to its comparatively rigid, peptidoglycan-rich outer surface, characteristic to Gram-positive bacteria. The presence of proteinaceous fibrils on strain HB slightly shielded the membrane fixed charges on HBC12.

Direct probing by atomic force microscopy of the cell surface softness of a fibrillated and nonfibrillated oral streptococcal strain.
In this paper, direct measurement by atomic force microscopy (AFM) of the cell surface softness of a fibrillated oral streptococcal strain Streptococcus salivarius HB and of a nonfibrillated strain S. salivarius HBC12 is presented, and the data interpretation is validated by comparison with results from independent techniques. Upon approach of the fibrillated strain in water, the AFM tip experi...

Background: Probiotics have attracted a great attention aiming to develop natural non-toxic antioxidants, because of their role in decreasing the risk of reactive oxygen species (ROS) accumulation. Objectives: The purpose of this study was to assess the antioxidant activity of a probiotic Streptococcus salivarius ssp thermophillus (St.sa) and to evaluate its protective effect against the ...
متن کاملThe influence of ionic strength on the adhesive bond stiffness of oral streptococci possessing different surface appendages as probed using AFM and QCM-D
Bacterial adhesion to surfaces poses threats to human-health, not always associated with adhering organisms, but often with their detachment causing contamination elsewhere. Bacterial adhesion mechanisms may not be valid for their detachment, known to proceed according to a visco-elastic mechanism. Here we aimed to investigate influences of ionic strength on the adhesive bond stiffness of two s...
متن کاملPrevention of streptococcal pharyngitis by anti-Streptococcus pyogenes bacteriocin-like inhibitory substances (BLIS) produced by Streptococcus salivarius.
BACKGROUND & OBJECTIVES Streptococcus salivarius is a numerically prominent member of the human oral microbiota that produces a variety of bacteriocin-like inhibitory substances (BLIS) having in vitro inhibitory activity against S. pyogenes. Our previous studies of S. salivarius isolates from children using a deferred antagonism BLIS production (P)-typing scheme showed that the 9 per cent of ch...
متن کاملSalivaricin A2 and the novel lantibiotic salivaricin B are encoded at adjacent loci on a 190-kilobase transmissible megaplasmid in the oral probiotic strain Streptococcus salivarius K12.
The commercial probiotic Streptococcus salivarius strain K12 is the prototype of those S. salivarius strains that are the most strongly inhibitory in a standardized test of streptococcal bacteriocin production and has been shown to produce the 2,368-Da salivaricin A2 (SalA2) and the 2,740-Da salivaricin B (SboB) lantibiotics. The previously uncharacterized SboB belongs to the type AII class of ...
